铁死亡(Ferroptosis)是一种依赖铁的细胞死亡形式,其驱动机制主要是由于细胞内抗氧化防御系统的失效,尤其是谷胱甘肽过氧化物酶4(GPX4)依赖的抗氧化体系出现问题。铁死亡的特点是细胞内过度累积活性氧和脂质过氧化物,最终导致因膜完整性丧失而发生的细胞死亡。
铁死亡在多种疾病中发挥着重要的作用,目前已证实在肝细胞癌、乳腺癌、黑色素瘤、卵巢癌和肾癌等不同类型的癌症中,铁死亡在限制癌细胞生长、增强细胞杀伤和克服耐药性方面发挥着关键作用。
